Hei!

Iīm doing my masterīs theses in the Bioprocess Engineering Department at the
University of Oulu. I want to repair a frameshift mutation in an E. coli gene
and it has been reported that the frameshift can be repaired by inserting 2
basepairs or deleting 1 bp in a 10bp region preceding the frameshift site.
Does anybody know how to get 2bp in there? I thought about homologous
recombination, phages,..? The problem is that I should not put a resistence
marker in (-> screening??). Are there any integration vectors I could use?
If sombody has experience with that, please contact me!
Thanks in advance,
           Christina
