Miniature sequencing devices by Oxford Nanopore Technologies (ONT) have made sequencing of DNA and RNA easier than ever before, but it is still complicated enough for beginners, especially data processing, analysis and interpretation. Although the sequencing devices are small (USB-stick or smartphone size), additional equipment and reagents are needed to prepare the DNA or RNA into sequenceable form, filling up at least a suitcase. Nevertheless, successful field sequencing has already been demonstrated for ONT devices. In the workshop, we'll demonstrate how to use ONT devices using both small lectures as well as lab demonstrations. We will outline how libraries are prepared, demonstrate the loading and sequencing using ONT’s MinION MK1C devise, go through the basics of monitoring sequencing progress, discuss the basic steps of basecalling (translation of raw signal into sequences) and demultiplexing, and then demonstrate the aspects of the data analyses.
