Abstract: The public is being told several myths about human reproduction and development that pretend to be science. The first myth is that all biologists agree that fertilization is when a separate human life begins. In reality, different groups of scientists have proposed that a separate human life begins at fertilization (acquisition of genome), gastrulation (acquisition of biological identity), EEG patterning (acquisition of the neural requisites for cognition and pain), viability and birth (the anatomical and physiological separation from the mother). And some scientists don't see this as a biological question. The second myth is that fertilization is solely a story of sperm performance. Indeed, the popular view of fertilization is patterned by the classical "myth of the hero." The scientific story of fertilization is one of remarkable interactions between the gametes and between the gametes and the female reproductive system. The third myth is that DNA is our "soul," our essence, and that our physical and behavioral phenotypes are derived almost exclusively from our genome. This has become a very popular view (as in Dawkins' writings), although the science behind it is very weak, and there are numerous counterexamples. These three myths inform and give support for each other's misinformation. The developmental biology concerning fertilization, phenotype production, and birth should be publicized more frequently to prevent the misuse of science for ideological and political concerns. Scott F. Gilbert is the Howard A. Schneiderman Professor of Biology, emeritus, at Swarthmore College. He is also a Finnish Distinguished Professor at the University of Helsinki. Professor Scott Gilbert is a renowned figure in the field of developmental biology. He has made significant contributions to our understanding of embryonic development and evolutionary biology. Gilbert's research into developmental biology often delves into how organisms adapt to different environments. This understanding is critical in the context of Planetary Health, where the goal is to protect both human and ecosystem health. In addition to his contributions in the field of developmental biology, Scott Gilbert is also known for his authorship of developmental biology textbooks. His textbooks have become widely recognized and utilized resources, serving as essential educational materials for students, researchers, and professionals interested in the subject. Thereby, the new therapy options and better patient stratification strategies for existing therapies are needed. Basal-like triple-negative (BL-TNBC) and HER2 positive (HER2+) breast cancers are among the clinically most aggressive subtypes. Whereas BL-TNBC patients lack any molecularly targeted therapies, HER2+ cancers can be targeted by therapeutic antibodies, and by small molecule HER2/EGFR inhibitors. However, a significant fraction of HER2+ breast cancers treated with these agents eventually relapse and develop to a deadly disease. In addition, both BL-TNBC and HER2+ cancers are relatively more resistant to both chemotherapy and radiation therapy than the other breast cancer types. Thereby, further understanding of the molecular mechanisms driving progression and therapy resistance of these aggressive breast cancer subtypes is a clinically highly relevant cancer research question. Phosphatases control all major oncogenic pathways and they have an important role in cancer therapy resistance. Phosphatases have also recently become druggable by small molecules. However, which specific phosphatases are particularly relevant as disease and therapy resistance drivers, or as therapy targets, in which breast cancer subtype is still poorly characterised. In this presentation, I will discuss our most recent findings identifying the roles or PP2A and DUSP6 phosphatases in BL-TNBC and HER2+ breast cancers, respectively, and present preclinical data indicating that their pharmacological targeting could provide a novel approach to overcome therapy resistance. The challenge this approach poses is made more complex by the numerous published definitions of welfare, which can lead to different interpretations of the same behavioral observation; the range of behavioral normalcy for a species or population of animal (i.e., its biology); individual animal differences; the time when behavioral observations are conducted; the appropriate weighting of any abnormalities observed; and the need to minimize any personal bias of the observer. A well-designed Behavioral Management Program (BMP) takes these considerations into account. The BMP is an overarching framework that aims to improve research animal welfare through consideration of social housing, environmental enrichment, animal training, facility design, and the assessment of behavior and mitigation of observed behavioral abnormalities. The various intrinsic and extrinsic factors that impact research animal behavior will be described, as these form the foundation of a successful BMP. When appropriately gathered and interpreted, behavior observations can be an important and practical tool to assess welfare and validate that management practices address not only extant behavioral concerns but the underlying welfare issue. SPEAKER Dr. Kathryn Bayne, CEO of AAALAC International, is a renowned veterinary applied animal behaviorist. She has led research programs on psychological well-being and environmental enrichment for various laboratory animal species at the NIH.
