CRC soluviljelijät kokous ma 29.5.2023 Yhteenveto 1. Huomioikaa, että kaikki soluviljelyyn tarvittavat/tilattavat materiaalit (vesi, viina, hanskat, sellu ja imupussit) jotka hankitaan Antin kautta tulee hoitaa ennen heinäkuun kesälomasesonkia. Antti on kesälomalla 3.-31.7. 2. Loma-ajan järjestelyjä: Nestetyppivuoro siirtyy Gynelle heinäkuun alussa, tarvittaessa apua on saatavilla myös muidenkin tutkimusryhmien työntekijöiden toimesta. Tekninen jaosto huolehtii inkubaattoreiden hiilidioksidipullojen vaihdon ja kaasun riittävyyden lomien aikana. Kantasolulaboratoriossa ei ole toimintaa kesällä ja sen vuoksi inkubaattorit sammutetaan. Muiden soluviljelylaboratorioiden puhtaudesta huolehditaan sopimalla ylläpidosta heinäkuussa töissä olevien kanssa. 3. Lomien jälkeen kaappien sterilointi kannattaa aikatauluttaa niin, että kaasumittaus voidaan tehdä samalla kertaa useammalle kaapille. 4. Corningin 10 ja 25 ml:n pipeteissä on huomattu valuvikaa. Hyvitystä varten viallinen pipetti tulisi ottaa talteen ja/tai kuvata. Laura on reklamoinut ongelmasta firmaan, häneen kannattaa olla yhteydessä jos viallisia pipettejä vielä ilmenee. 5. Keskusteltiin steriilistä työskentelystä ja siihen liittyvistä vaihtelevista käytänteistä. Todettiin, että laminaareissa kaikkien yleiseen käyttöön (pasteur pipettien noukkimiseen) tarkoitetut pinsetit eivät ole puhtausasteeltaan soveltuvia solujen kanssa suoraan kosketuksissa oleviin pintoihin. Esim. peitinlasien asetteluun maljalle kannattaa käyttää vain omaan käyttöön steriloituja pinsettejä. 6. Pipettien huollosta ja kantasolulaboratorion imusysteemistä keskusteltiin. Pipettien käyttöikä on rajallinen ja akkua vaihtamalla käyttöä voidaan jatkaa hetkellisesti kohtuullisin kuluin. Pipetit on myös hyvä lähettää huoltoon tarpeen vaatiessa. Pipettejä on huollettu viimeksi 2018 ja 2020. Jos tarvista, näitä pipettihuoltoja voisi aloittaa lomien jälkeen. 7. Anja tekee mykoplasmatestin ma 5.6. Kitin toimituksessa on ollut hankaluuksia ja viiveitä, toimittajaa vaihdetaan. Jatkossa kitti tilataan City-Labin kautta. 8. Työtakit viedään pesuun ti 27.6. joten tyhjentäkää takkienne taskut ennen sitä. 9. Seuraava kokous ma 4.9.23 klo 13.30 CRC:n kahvihuone.

Summary 1. Please note that all materials needed/ordered for cell culture (water, ethanol, gloves, tissue and bags) that are obtained through Anti must be taken care of before the July summer holiday season. Antti is on summer vacation from July 3-31. 2. Arrangements for vacation time: The liquid nitrogen shift will be transferred to Gyne at the beginning of July, if necessary, help is also available from other research groups. The technical department takes care of changing the incubators' carbon dioxide bottles and ensuring that there is enough gas during the holidays. The stem cell laboratory does not operate in the summer and therefore the incubators are turned off. The cleanliness of the other cell culture laboratories is taken care of by agreeing on maintenance in July with those working. 3. After the holidays, you should schedule the sterilization of the incubators so that the gas measurement can be done for several cases at the same time. 4. Problems has been noticed in Corning's 10 and 25 ml pipettes. For a refund, pipettes with flaws should be restored and/or photographed. Laura has complained about the problem to the company, you should contact her if problems still appear. 5. Sterile working and related varying practices were discussed. It was found that the tweezers intended for general use in laminars (for picking up Pasteur pipettes) are not sterile enough for surfaces in direct contact with cells. For example, to place the coverslips on the dish, please, use sterilized tweezers intended for your own use. 6. The maintenance of pipettes and the suction system of the stem cell laboratory were discussed. Pipettes have a limited lifespan, and by changing the battery, use can be temporarily continued at reasonable costs. Pipettes can be sent for maintenance if necessary, for example after the holidays. 7. Anja will do a mycoplasma test on Mon 5.6. There have been difficulties and delays in the delivery of the kit, the supplier is being changed. In the future, the kit will be ordered through City-Lab. 8. The lab coats will be taken to the wash on Tuesday 27.6. so empty the pockets before then. 9. Next meeting Mon 4.9.23 at 1:30 p.m. CRC coffee room.
