Soluviljelijöiden kokous ke 28.8. CRC:n kahvihuone
1. Kokouksessa keskusteltiin mykoplasmasta ja mietittiin ohjeistusta, kuinka vältämme jatkossakin kontaminaation leviämisen soluviljelmiimme:
Aika ajoin tutkimusryhmille lähetetään solulinjoja ulkopuolisilta kollaboraattoreilta. Näissä tilanteissa on tärkeää pyytää mykoplasmatestaus ennen lähetystä ja ottaa vastaan vain negatiiviset solulinjat. Jos testaus ei jostain syystä ole mahdollista ennen lähetystä ja solut lähetetään pakastettuina, kannattaa pyytää pieni määrä kasvatusmediumia erikseen pakastettuna, jotta voimme tehdä CRC:llä mykoplasmatestin ennen solujen sulattamista kasvatukseen. Lisäksi olemme hankkimassa CRC:lle Cell Locker isolaattoria https://www.fishersci.be/shop/products/cell-locker/15864285, joka laitetaan inkubaattorin sisään ja mikä eristää sisällään olevat solut ja estää mahdollisten patogeenien leviämisen muualle inkubaattoriin. Kaasut pääsevät kiertämään isolaattorissa filtterin läpi. Isolaattoria voi käyttää tilanteessa, jossa mykoplasmaa ei ole vielä poissuljettu viljelmästä.
Seuraava mykoplasmatesti tullaan tekemään 30.9.2024. Näytteitä voi tuoda labraan 377B 30.9. klo 9 mennessä.
2. Solulaskurin toiminnasta oli keskustelua, mutta vaikutti, että laite toimii tällä hetkellä eikä ole tarvetta sen kalibrointiin liittyviin suht tyyriisiin hankintoihin.
3. Soluviljelylaminaareihin hankitaan telineet joihin voi virittää roskapussin. Tämä on käytännöllisempää tilanteisiin, jossa soluviljelyyn liittyvää muoviroskaa kertyy paljon eikä perinteiset pienet muovikaukalot riitä.
4. Seuraava kokous pidetään to 10.10. klo 13 CRC:n kahvihuoneessa.
Cell culture meeting Wed 28.8. CRC coffee room
1. In the meeting, we discussed mycoplasma and the guidelines for how we can continue avoiding the contamination in our cell cultures: From time to time, cell lines are sent to research groups from external collaborators. In these situations, it is important to request mycoplasma testing before shipment and only accept negative cell lines. If for some reason testing is not possible before shipment and the cells are sent frozen, it is recommended to request a small aliquot of culture medium separately frozen so that we can perform a mycoplasma test with CRC before thawing the cells for culture. In addition, we are getting a Cell Locker isolator https://www.fishersci.be/shop/products/cell-locker/15864285 for CRC, which can be placed inside the incubator and which isolates the cells inside and prevents possible pathogens from spreading to the rest of the incubator. The gases can circulate in the insulator through the filter. The isolator can be used in situations where mycoplasma has not yet been excluded from the culture.
The next mycoplasma test will be done on September 30, 2024. Samples can be brought to lab 377B on September 30 by 9 o'clock.
2. There was a discussion about the operation of the cell counter, but it seemed that the device is working at the moment and there is no need for relatively expensive purchases related to its calibration.
3. Stands will be purchased for the cell culture laminars, on which a garbage bag can be attached. This is more practical for situations when there is a lot of plastic waste and traditional small plastic trays are not enough.
4. The next meeting will be held on Thursday, October 10 at 1 pm in the CRC coffee room.
