Soluviljelijöiden kokous ti 3.12.

CRC:n kahvihuone

1. Anja tekee mykoplasmatestin ma 9.12. Näytteitä voi toimittaa hänelle labraan 377B vielä maanantainaamuun klo 9 mennessä.

2. Juulia on kirjoittanut seikkaperäiset toimintaohjeet mykoplasmaepäiltyjen kudos- ja solunäytteiden käsittelyyn. Ohjeet on tallennettu yhteiseen kansioon:

3. Antti on tiedustellut sairaalasta mahdollisesti muuton vuoksi käyttämättömäksi jääviä imusysteemejä meille soluviljelyyn. Yhteyshenkilön mukaan laitteita ei tällä hetkellä ollut, mutta hän ilmoittaa jos niitä ilmenee.

4. IPSilän toinen inkubaattori on ollut useamman kuukauden käyttämättömänä ja nyt avatessa tuntuu homeen haju. Kaappi on jo yli 20 vuotta vanha, eikä siinä ole sterilointiohjelmaa. Keskusteltiin keinoista joilla kaapin voisi tehokkaasti puhdistaa (esim. biosidal, etanoli). Kannattaa myös tarkistaa valmistajan sivuilta ohjeet puhdistukseen. Kaappiin laitetaan joulukuussa testikasvatus jonka avulla seurataan ilmestyykö kontaminaatiota tai vaikuttaako kaapin olosuhteet muuten kasvuun. Yleisenä ohjeena, kannattaa sammuttaa kaappi (ja poistaa vesiastia) jos siellä ei viljellä soluja pariin kuukauteen.

5. Nestetyppitankkien lähestyvästä tilanpuutteesta keskusteltiin. Nestetyppitankkihuoneessa ei ole enää tilaa uudelle tankille vaan säilytykseen täytyy miettiä muita ratkaisuja. Yksi esille tullut vaihtoehto on ultrasyväjääpakastin (~150C) jossa arkistoitavia solulinjoja voitaisiin säilyttää tarvittaessa myös CRC:n ulkopuolella.

6. Lähestyvään joulutaukoon valmistaudutaan seuraavasti. 341/2B labroista yksi kaappi puhdistetaan ja kaasut tarkistetaan jo ennen joulutaukoa niin että viljely voidaan aloittaa heti joulun jälkeen. Toiset kaapit puolestaan puhdistetaan ensi vuoden alussa tammikuun ensimmäisellä viikolla. Takit viedään pesuun ma 16.12. iltapäivällä. Nestetypen kuljetuksen aikataulut selvitetään ensi viikon CRC:n labrapalaveriin, jossa käydään läpi toimintasuunnitelma tankkien täytön suhteen.

7. Seuraava kokous pidetään ke 5.2. klo 13 CRC:n kahvihuoneessa.

Cell culture meeting Tue 3.12.

CRC coffee room

1. Anja will do a mycoplasma test on Monday 9 December. Samples can be delivered to her in lab 377B by 9 a.m. on Monday morning.

2. Juulia has written detailed operating instructions for handling tissue and cell samples suspected of mycoplasma. The instructions can be found from a shared folder:

3. Suction systems left unused due to the move have been inquired from the hospital. According to the contact person, there were no devices at the moment, but he will let us know if they appear.

4. One of the incubators in the IPSC lab has been unused for several months and now when opened, the smell of mold is evident. The incubator is already more than 20 years old, and it does not have a sterilization program. We discussed ways to effectively clean the incubator (e.g. biocidal, ethanol). The instructions for cleaning on the manufacturer's website should be also checked. In December, test growth will be put in the incubator, which will be used to monitor whether contamination appears or whether the conditions in the cabinet otherwise affect the growth. As a general guideline, it is good to turn off the incubator (and remove the water container) if you do not cultivate cells there for a couple of months.

5. The lack of space in liquid nitrogen tanks is predictable. There is no more room for a new tank in the liquid nitrogen tank room, so other solutions for storage are needed. One option that came up is an ultra-deep freezer (~150C) where cell lines to be archived could also be stored outside the CRC if necessary.

6. Preparations for the approaching Christmas break: One incubator from both labs (341/2B) is cleaned and the gases are checked already before the Christmas break, so that culturing can start immediately after Christmas. The other incubators will be cleaned at the beginning of next year, in the first week of January. The coats will be taken to the wash on Mon 16.12. afternoon. The schedules for the transport of liquid nitrogen will be clarified at next week's CRC lab meeting.

7. The next meeting will be held on Wed 5.2. at 1 pm in the CRC coffee room.
