Hypoxia-inducible factor (HIF) prolyl-4-hydroxylases (HIF-P4Hs) isoenzymes are oxygen sensors that hydroxylate the HIF-α subunit under normoxia and are implicated in the mechanism of the organism adaptation to low oxygen levels. Three HIF-P4H isoenzymes are known in mammals: HIF-P4Hs 1, 2, and 3. A fourth P4H (P4H-TM) that contains an endoplasmic reticulum transmembrane domain was recently identified by our group (Koivunen et al. 2007) and it is challenge now to characterize it’s function. This pro gradu project will be about the roles of HIF-P4Hs in the brain, in particular in stroke disease. This project is heavily based on use of different transgenic mouse models. Methods will include primary cell culture of different neural cell types isolated from mouse embryos, RNA extraction from cells and mouse tissue,  quantitative PCR, Western blot, immunostaining, various functional assays on synaptic plasticity and cell death, etc.
