There is a pro gradu project available in Prof. Johanna Myllyharju’s group at the Department of Medical Biochemistry and Molecular Biology. The starting date of pro gradu is flexible (from August
till October 2015) and can be negotiated.
The research of the group is focused on the role of Hypoxia-inducible factor (HIF) prolyl-4-hydroxylases (HIF-P4Hs) isoenzymes (
http://www.oulu.fi/biocenter/groups/myllyharju). These are oxygen sensors that regulate organism adaptation to low oxygen levels.
The proposed pro gradu project will be about the roles of HIF-P4Hs in the nervous system. It will be focused on link between HIF-P4Hs and calcium signaling on one hand and on the functional discrepancy in neural cells caused by changes in calcium signaling
on the other hand. This project is heavily based on use of different transgenic mouse models. In the moment we have knockout/
hypomorphic mice available for each of the four isoenzymes existing in mammals (HIF-P4Hs 1, 2, 3 and HIF-P4H TM). Methods will include primary cell culture of different neural cell types isolated from newborn mice, RNA/protein
extraction from cells and mouse tissues, quantitative PCR, Western blot, immunostaining, cryosectioning & histology, calcium imaging, various functional assays on synaptic plasticity and cell death, usage of pharmacological agonists/antagonists.
