Soluviljelijöiden kokous pe 15.12.2023

Yhteenveto

1. Edelleen hiilidioksidipullot 341B ja 342B soluviljelyjen yhteydessä tyhjenevät aiempaa nopeammin ja mahdollista kaasuvuotoa selvitetään. Kaasupullot käydään vaihtamassa ennen joulutaukoa viikolla 51 jotta vältytään hälytykseltä joululoman aikaan. Vuoden vaihteen jälkeen ti 2.1. tekniset palvelut aloittavat 342B laboratorion inkubaattoreiden toiminnan testauksella. Kaapit testataan samanaikaisesti jolloin molemmat tulee olla tyhjillään soluista. Viljelyssä olevat solut siirretään siksi aikaa 342B laboratorion inkubaattoriin. Tavoitteena on saada myös 341B:n kaapit tarkistettua yksi kerrallaan ensimmäisen viikon aikana, ennen kuin soluviljelytoiminta aktivoituu taas vuoden vaihteen jälkeen. Kaappien steriloinnit tehdään tarkistusten jälkeen, hyllyjä voi laittaa pesuun jo etukäteen jos kaappi on suljettu jouluksi. Huoneilman hiilidioksidipitoisuuden mittaukset aloitettiin 357B tilasta. Tuloksena huomattiin, että hiilidioksipitoisuus saattaa nousta yli raja-arvon viikonloppuna jolloin ilmanvaihto koko rakennuksessa on pienemmällä. Kannattaa siis huolehtia riittävästä ilmanvaihdosta avaamalla eteistilan ovi, jos työskentelee viikonloppuna 357B tilassa. 341B ja 342B tilojen hiilidioksidipitoisuudet mitataan vuodenvaihteen jälkeen. Kannattaa kiinnittää erityistä huomiota taukoihin sekä olotilan muutoksiin, jos työskentelee viikonloppuisin näissä tiloissa. Teknisiä palveluita sekä laboratoriopäällikköä on tiedotettu asiasta, tieto viedään myös työsuojeluvaltuutetulle.

2. Nestetyppitankkien täytöstä ja nestetypen tarpeesta joululoman aikaan keskusteltiin. Sovittiin, että Maarit peruu täytön välipäiviltä, mutta ensimmäisellä viikolla tankkien täyttö on taas tarpeen. Gynen Riikka ja Ulla huolehtivat nestetyppitankkien täytöstä ensimmäisellä viikolla. Sen jälkeen nestetyppivuorossa on Neuro.

3. Labratakit viedään pesuun maanantaina 18.12. iltapäivällä. Tyhjentäkää takkienne taskut, takit laitetaan 341/342B eteistilassa olevaan säkkiin. Tarkistakaa samalla soluviljelyssä käytössä olevat kengät. Jätetään eteistilaan ainoastaan aktiivisessa käytössä olevat ja käyttämättömät tai harvoin tarvittavat viedään jätehuoneessa olevaan laatikkoon josta ne voi hakea käyttöön jos/kun tarpeen.

4. Mykoplasmanäytteet ehtii vielä toimittaa Anjalle, jos on nopea! Anja ottaa testiin mukaan viimeistään ma 18.12. klo 9 mennessä toimitetut näytteet.

5. Hankinnoista keskusteltiin. Pipetti tms. roskien keräämiseen laminaareissa tilataan kertakäyttöpussit ja niille telineet. Selvitellään myös mikroskoopin hintaa jossa parempi kuvantamismahdollisuus, tosin rahoitusta hankintaan ei tällä hetkellä ole.

6. Yleiset ohjeet, vastuuhenkilöt ja yhteyshenkilöt päivitettiin. Tallennan ohjeet tämän viikon aikana kansioon: S:\KLIIN\CRC\Soluviljely Cell culture\Instructions . Päivittäkää ohjeet myös printattuna kansioihin jotka löytyvät kolmesta soluviljelylabrasta ja muistakaa ohjeistaa uusia työntekijöitä tutustumaan niin ennen soluviljelyn aloittamista.

Cell culture meeting Fri 15.12.2023

Summary

1. Carbon dioxide bottles connected to incubators in 341B and 342B continue emptying faster than before and a possible gas leak is investigated. The gas bottles are changed before the Christmas break in week 51 to avoid an alarm during the Christmas holidays. After the new year, Tue 2.1. technical services start testing the operation of incubators in the 342B laboratory. Both incubators are tested at the same time, so both of them must be empty of cells. The cells in culture are therefore transferred to the 342B laboratory incubator. The goal is to also have the incubators of 341B checked one at a time during the first week, before cell cultures are activated again. Incubators are sterilized after the thorough check, shelves can be washed in advance if the incubators are closed for Christmas. The measurements of the carbon dioxide in the room air were started in room 357B. As a result, it was noticed that the level of carbon dioxide may rise just above the limit value during the weekend, when the ventilation in the entire building is lower. It is therefore worth taking care of sufficient ventilation by opening the entrance door if you work in room 357B on the weekend. The carbon dioxide levels in the room air in premises 341B and 342B are measured after the new year. You should pay special attention to breaks and general condition if you work on weekends in these spaces. Technical services and the laboratory manager have been informed about the matter, the information will also be taken to the occupational health and safety representative.

2. The filling of liquid nitrogen tanks and the need for liquid nitrogen during Christmas time were discussed. It was agreed that Maarit will cancel filling in week 52, but in the first week of 2024 it will be necessary to fill the tanks again. Gyne's Riikka and Ulla take care of filling the liquid nitrogen tanks in the first week. After that, it's Neuro on the liquid nitrogen shift.

3. Lab coats will be taken for washing on Monday 18.12. afternoon. Empty the pockets of your coat and put them in the sack in the entrance room of 341/342B. At the same time, check the shoes used in cell culture. Only those that are in active use are left in the rack, and those that are not used or rarely needed are taken to the box in the waste room from where they can be picked up when/if needed.

4. There is still time to deliver samples for mycoplasma test to Anja, if you hurry! Anja accepts samples until 9 am today (Monday 18th December).

5. Equipment investment needs were discussed. Disposable bags and racks are ordered for collecting pipettes, etc. garbage in laminars. We are also investigating the price of a microscope with better imaging capabilities, although there is currently no funding for the purchase.

6. General instructions, responsible persons and contact persons were updated. The instructions can be found in electronic form from the folder later this week: S:\KLIIN\CRC\Soluviljely Cell culture\Instructions. Please also update the printed instructions to the folders that can be found in the three cell culture labs and remember to instruct new employees to familiarize themselves with it before starting cell culture.
