Soluviljelijöiden kokous to 10.10. CRC:n kahvihuone
1. Cell Locker isolaattori on toimitettu ja kokouksessa mietittiin sen käyttöperiaatetta. Todettiin, että käytetään isolaattoria testaamattomille soluille jotka lähetetty CRC:lle muualta (kansainvälisestä) tutkimuslaboratorioista. Jatkossakin potilaiden kudosbiopsioista perustetut primääriviljelmät voidaan kasvattaa suoraan inkubaattoreissa ilman isolaattoria kunhan huolehditaan mykoplasman testaamisesta niin pian kuin mahdollista.
2. Anjan kanssa keskusteltiin säännöllisen, aiempaa tiuhempaan suoritettavan mykoplasmatestauksen tekemisestä. Testien väli olisi noin kerran parissa kuukaudessa riippuen kertyvästä näytemäärästä. Tätä varten on perustettu sähköinen lomake kansioon:

jonne jokainen voi merkitä testattavat näytteet. Lomaketta voi täydentää, jos nyt huomaatte sieltä jonkin olennaisen tiedon puuttuvan. Kun listalle on kertynyt seuraavat 17 näytettä, Anja tekee testin.
3. Soluviljelyn 341B inkubaattoreissa on tällä hetkellä ruuhkaa ja kokouksessa keskusteltiin mahdollisesta ryhmien uudelleen järjestäytymisestä eri soluviljelytiloihin. Muuton suhteen haasteena on rajallinen hylly- ja säilytystila 342B:ssa mikä hankaloittaa isomman ryhmän sinne siirtymistä. 342B:n inkubaattorit ovat tällä hetkellä vajaalla käytöllä joten niihin voi viedä solut (sekä Cell Locker isolaattorin) vaikka viljely tapahtuisikin 341B:n laminaarissa. Kannattaa kuitenkin huomioida ettei kaapia ole hyvä vaihtaa kesken kokeen. Varauslistat molempien laboratorioiden laminaareihin olisi hyvä avata näissä kahdessa tilassa työskenteleville tutkijoille.
4. Labran siisteyden haasteista keskusteltiin. On tärkeää, että kaikki huolehtivat lähtiessään labran siistiin kuntoon seuraavalle työskentelijälle. Tämä käsittää mm. imulaitteiden huolellisen puhdistamisen, pullojen ja käyttöliuosten täyttämisen ja myös osallistumisen säännölliseen laboratorion ylläpitoon sekä roskien ja tiskien viemiseen autoklavoitavaksi. Uusien työntekijöiden perehdyttämisen tärkeydestä soluviljelylaboratorion toimintaperiaatteisiin keskusteltiin. Mietittiin, olisiko tarpeen laatia kirjallinen ohjeistus perehdytykseen sekä kuvat havainnollistamaan puhtaanapitoa. Perehdytys toteutetaan jokaisessa ryhmässä vastaavan henkilön toimesta mutta todettiin, että jokainen voi ystävällisesti opastaa muita työntekijöitä jos huomaa toimintaperiaatteissa korjattavaa.
5. Seuraava kokous pidetään ti 3.12. klo 13 CRC:n kahvihuoneessa.
Cell culture meeting to 10.10. CRC coffee room
1. The Cell Locker isolator has been delivered and the principle of its use was discussed at the meeting. It was concluded that the isolator should be used for untested cells sent to CRC from other (international) research laboratories. In the future, primary cultures established from patient tissue biopsies can be grown in incubators without an isolator, as long as care is taken to test for mycoplasma as soon as possible.
2. We discussed with Anja about doing regular, more frequent mycoplasma testing. The interval between the tests would be about once every couple of months, depending on the amount of samples collected. For this purpose, an electronic form has been created in the folder:
S:\KLIIN\CRC\Cell culture Cell culture
where everyone can mark the samples to be tested. You can complete the form if you now notice that some essential information is missing. When the next 17 samples have been listed, Anja does the test.
3. The cell culture 341B incubators are currently overcrowded, and we discussed the possible reorganization of groups into different cell culture facilities in the meeting. The challenge regarding the move is the limited shelf and storage space in 342B, which makes it difficult for a larger group to move there. The incubators of 342B are currently underutilized, so cells (and the Cell Locker isolator) can be taken into them, even if using laminars of 341B. However, it is worth noting that it is not good to change the incubator in the middle of the experiment. It would be good to open the reservation lists for laminars in both laboratories to researchers working in these two facilities.
4. The challenges of keeping the lab clean were discussed. It is important that everyone ensures that the lab is in a clean condition for the next user when they leave. This includes e.g. cleaning the suction equipment carefully, filling bottles and working solutions, and also participating in regular laboratory maintenance duties including the transportation of garbage and dishes to be autoclaved. The importance of introducing new employees to the operating principles of the cell culture laboratory was discussed. We thought about whether it would be even necessary to prepare written instructions for orientation and pictures to illustrate cleaning. Orientation is generally carried out by the responsible person in each research group, but it was stated that everyone can kindly guide others if they notice something to be corrected in their operating principles.

Soluviljelijöiden kokous ti 3.12. CRC:n kahvihuone
1. Anja tekee mykoplasmatestin ma 9.12. Näytteitä voi toimittaa hänelle labraan 377B vielä maanantainaamuun klo 9 mennessä.
2. Juulia on kirjoittanut seikkaperäiset toimintaohjeet mykoplasmaepäiltyjen kudos- ja solunäytteiden käsittelyyn. Ohjeet on tallennettu yhteiseen kansioon: S:\KLIIN\CRC\Soluviljely Cell culture\Instructions
3. Antti on tiedustellut sairaalasta mahdollisesti muuton vuoksi käyttämättömäksi jääviä imusysteemejä meille soluviljelyyn. Yhteyshenkilön mukaan laitteita ei tällä hetkellä ollut, mutta hän ilmoittaa jos niitä ilmenee.
4. IPSilän toinen inkubaattori on ollut useamman kuukauden käyttämättömänä ja nyt avatessa tuntuu homeen haju. Kaappi on jo yli 20 vuotta vanha, eikä siinä ole sterilointiohjelmaa. Keskusteltiin keinoista joilla kaapin voisi tehokkaasti puhdistaa (esim. biosidal, etanoli). Kannattaa myös tarkistaa valmistajan sivuilta ohjeet puhdistukseen. Kaappiin laitetaan joulukuussa testikasvatus jonka avulla seurataan ilmestyykö kontaminaatiota tai vaikuttaako kaapin olosuhteet muuten kasvuun. Yleisenä ohjeena, kannattaa sammuttaa kaappi (ja poistaa vesiastia) jos siellä ei viljellä soluja pariin kuukauteen.
5. Nestetyppitankkien lähestyvästä tilanpuutteesta keskusteltiin. Nestetyppitankkihuoneessa ei ole enää tilaa uudelle tankille vaan säilytykseen täytyy miettiä muita ratkaisuja. Yksi esille tullut vaihtoehto on ultrasyväjääpakastin (~150C) jossa arkistoitavia solulinjoja voitaisiin säilyttää tarvittaessa myös CRC:n ulkopuolella.
6. Lähestyvään joulutaukoon valmistaudutaan seuraavasti. 341/2B labroista yksi kaappi puhdistetaan ja kaasut tarkistetaan jo ennen joulutaukoa niin että viljely voidaan aloittaa heti joulun jälkeen. Toiset kaapit puolestaan puhdistetaan ensi vuoden alussa tammikuun ensimmäisellä viikolla. Takit viedään pesuun ma 16.12. iltapäivällä. Nestetypen kuljetuksen aikataulut selvitetään ensi viikon CRC:n labrapalaveriin, jossa käydään läpi toimintasuunnitelma tankkien täytön suhteen.

Cell culture meeting Tue 3.12. CRC coffee room
1. Anja will do a mycoplasma test on Monday 9 December. Samples can be delivered to her in lab 377B by 9 a.m. on Monday morning.
2. Juulia has written detailed operating instructions for handling tissue and cell samples suspected of mycoplasma. The instructions can be found from a shared folder: S:\KLIIN\CRC\Cell culture Cell culture\Instructions
3. Suction systems left unused due to the move have been inquired from the hospital. According to the contact person, there were no devices at the moment, but he will let us know if they appear.
4. One of the incubators in the IPSC lab has been unused for several months and now when opened, the smell of mold is evident. The incubator is already more than 20 years old, and it does not have a sterilization program. We discussed ways to effectively clean the incubator (e.g. biocidal, ethanol). The instructions for cleaning on the manufacturer's website should be also checked. In December, test growth will be put in the incubator, which will be used to monitor whether contamination appears or whether the conditions in the cabinet otherwise affect the growth. As a general guideline, it is good to turn off the incubator (and remove the water container) if you do not cultivate cells there for a couple of months. 5. The lack of space in liquid nitrogen tanks is predictable. There is no more room for a new tank in the liquid nitrogen tank room, so other solutions for storage are needed. One option that came up is an ultra-deep freezer (~150C) where cell lines to be archived could also be stored outside the CRC if necessary.
6. Preparations for the approaching Christmas break: One incubator from both labs (341/2B) is cleaned and the gases are checked already before the Christmas break, so that culturing can start immediately after Christmas. The other incubators will be cleaned at the beginning of next year, in the first week of January. The coats will be taken to the wash on Mon 16.12. afternoon. The schedules for the transport of liquid nitrogen will be clarified at next week's CRC lab meeting.

Soluviljelijöiden kokous ke 5.2. CRC:n kahvihuone
1. Anja teki mykoplasmatestin joulukuussa. Kaksi positiivista näytettä löytyi jotka tuhottiin heti tuloksen selvittyä asianmukaisesti. Näytteet olivat lähtökohtaisesti jo mykoplasmaepäiltyjä. Sen vuoksi niitä ei käsitelty soluviljelylaboratoriossamme lainkaan vaan niiden kanssa toimittiin alusta saakka erityisellä huolellisuudella ja varovaisuudella noudattaen toimintaohjeitamme: S:\KLIIN\CRC\Soluviljely Cell culture\Instructions
2. Cell Locker on todettu toimivaksi ratkaisuksi ulkopuolelta tulleiden solulinjojen karanteeniin ennen kuin ne on todettu mykoplasma negatiivisiksi. Lockeria on pidetty inkubaattorissa 342B labrassa. Tämän kevään aikana on tulossa ruuhkaa kaappeihin ja sen vuoksi isolaattori täytyy siirtää pois kaapista. Pohdimme mahdollisuutta säilyttää isolaattori puhtaana autoklavointipussissa inkubaattorin ulkopuolella.
3. Imulaitteita on nyt saatu sairaalan muuton yhteydessä jokaiseen soluviljelylaboratorioon ja niiden käyttöä ajetaan parhaillaan sisään. Kiitos Antille asian hoitamisesta!
4. IPSilän toinen inkubaattori lakkasi toimimasta ja sen tilalle saatiin hankittua uusi Binder. Kaappi on saatu toimintaan.
5. Nestetyppitankkien vääjäämättä lähestyvästä tilanpuutteesta keskusteltiin ja sovittiin, että jokaisessa ryhmässä on tärkeää käydä läpi säilytyksessä olevat soluampullit. Olennaista olisi saada yhteys tutkijoihin jotka eivät tee enää aktiivisesti tutkimusta ja käydä heidän nimissään olevat näytteet läpi. Helpoin on aloittaa sähköisistä listoista ja sitä kautta selvittää kuinka paljon tilaa nyt vievät vanhat näytteet joita arkistoidaan ja joiden tarpeellisuudesta ei ole kenelläkään tietoa/käynnissä olevaa tutkimuslupaa.
6. Helmi-maaliskuun vaihteessa viikoilla 9, 10 ja 11 on suunnitelmissa iso soluviljelykoe joka tulee viemään suuren osan inkubaattorikapasiteetista soluviljelylaboratorioissa 341/2B. Keskusteltiin kokoukseen osallistujien kesken yleisestä tilantarpeesta tuona aikana, ja vaikuttaa että soluviljelytoiminta saadaan kyllä sumplittua eri ryhmien kesken. Ilmoitelkaa kuitenkin Saijalle ja Pirjolle, jos teillä on soluviljelyä suunnitteilla niinä viikkoina, etenkin jos ette olleet mukana kokouksessa. Parempi suunnitella porukalla ja varmistaa, että kaikkien soluviljely saadaan toteutettua jolloin vältytään ikäviltä viime hetken yllätyksiltä.
7. Kokouksessa keskusteltiin pitkään myös yleisestä laboratorio- ja soluviljelytilojen siisteyden ylläpidosta mihin kuuluu tiskien ja autoklaaviin menevien liuosten sekä jätteiden edestakainen kuljetus BMTK:n päässä sijaitsevaan välinehuoltoon sekä samalla CityLabiin saapuvien CRC:n ryhmien tilausten toimittaminen. Yhteisiin tehtäviin kuuluu myös nestetyppivuorot jotka pyörivät eri tutkimusryhmissä säännöllisin väliajoin. On erittäin tärkeää, että jokainen tutkimusryhmä osallistuu näihin yhteisöllisiin ylläpitotehtäviin. Jotta emme kuormita kohtuuttomasti yksittäisiä työntekijöitä, kannattaa suunnitella kiertävät vuorot myös tutkimusryhmän sisällä.
8. Seuraava kokous pidetään ti 1.4. klo 9 CRC:n kahvihuoneessa.
Cell culture meeting Wed 5.2. CRC coffee room 1. Anja did a mycoplasma test in December. Two positive samples were found, which were destroyed properly as soon as the results were determined. The samples were initially suspected of mycoplasma. Therefore, they were not brought to our cell culture laboratory at all, but were handled with special care and caution from the beginning, following our operating instructions: S:\KLIIN\CRC\Soluviljely Cell culture\Instructions
2. Cell Locker has been found to be a practical solution for quarantining arriving cell lines before they are tested for mycoplasma. Locker has been kept in the incubator of the laboratory 342B. During this spring, there will be a lot of cell culturing and thus limited space available in the incubators. Therefore, the isolator must be moved out of the incubator. In order to keep the filters clean, we are considering of storing the Cell Locker in an autoclave bag when not in use.
3. Suction systems left unused from the old hospital have now been received to each cell culture laboratory and their use is currently being initiated. Thanks to Antti for taking care of it!
4. The old incubator of IPS cell lab stopped working and a new Binder was purchased to replace it. The incubator is now in operation.
5. The lack of space in the liquid nitrogen tanks was discussed and it was agreed that it is important to go through the cell ampoules in storage in each group. It would be essential to get in touch with researchers who are no longer actively doing research and go through the samples in their names. It is easiest to start with electronic lists and thereby find out how much space is now taken up by the old samples that are archived and for which no one has any information about the necessity/current research permit.
6. Between February-March, in weeks 9, 10 and 11, massive cell culture experiment will take place, which will take up a large part of the incubator capacity in cell culture laboratories 341/2B. The meeting participants discussed the general need for space at that time, and it seems that the cell culture activities of different groups can be managed during the busy weeks. However, please inform Saija and Pirjo if you have cell culturing planned during those weeks, especially if you were not in the meeting. It is better to plan together and ensure that everyone's cell culture can be carried out, thus avoiding unpleasant last-minute surprises.
7. We also discussed general maintenance of laboratory and cell culture facilities, which includes the transport of dishes and autoclaved solutions and waste to the DWA unit located at the other end of the campus, FBMM, as well as delivering the orders of the CRC groups arriving at CityLab at the same time. Joint tasks also include liquid nitrogen shifts that rotate in different research groups at regular intervals. It is very important that each research group participates in these communal maintenance tasks. In order not to unreasonably burden individual employees, it is worth planning rotating shifts within the research groups as well.

CRC:n kahvihuone Kokouksessa keskusteltiin seuraavaa 1. Helmi-maaliskuun vaihteen ruuhkista on selvitty eikä ongelmia ole ilmennyt vaan tilaa on riittänyt kaikille. 2. Cell Lockerille on löytynyt riittävästi tilaa inkubaattorissa eikä sen säilytykseen kaapin ulkopuolella ole ollut tarvetta. Filtterit sovittiin vaihdettavaksi aina kaappien pesun yhteydessä eli juhannuksen ja joulun aikaan sekä jos ilmenee kontaminaatioita. 3. Nestetyppitankkien vääjäämättä lähestyvästä tilanpuutteesta keskusteltiin lyhyesti ja muistutettiin, että jokaisessa ryhmässä on tärkeää käydä läpi säilytyksessä olevat soluampullit. Olennaista olisi saada yhteys tutkijoihin jotka eivät tee enää aktiivisesti tutkimusta ja käydä heidän nimissään olevat näytteet läpi. Helpoin on aloittaa sähköisistä listoista ja sitä kautta selvittää kuinka paljon tilaa nyt vievät vanhat näytteet joita arkistoidaan ja joiden tarpeellisuudesta ei ole kenelläkään tietoa/käynnissä olevaa tutkimuslupaa. 4. Soluviljelystä tulevaa autoklavoitavaa jätettä sisältävät pussit sankkoineen siirretään soluviljelyjen etuhuoneeseen odottamaan noutoa. Pussit nidotaan valmiiksi kiinni ja merkitään onko kyseessä märkä vai kuiva jäte. Sen jälkeen soluviljelyyn viedään tyhjä sankko pusseineen soluviljelyn eteisestä tai kylmähuoneesta. Jätteet kerätään autoklavoitavaksi kolmena päivänä viikossa (ma, ke ja pe). Jos jätettä kertyy enemmän, silloin jokaisen on itse huolehdittava jätteet autoklavoitavaksi. 6. Nestetyppitankeille on hankittu uudet hanskat sekä säilytyslaatikkoja. 7. Haravapipettien kalibrointi on ajankohtaista. Pari pipettiä on ollut huollettavana, ilmoitelkaa Antille jos huomaatte lisää pipettejä joiden kalibrointi on aiheellista. 8. Seuraava kokous pidetään ke 4.6. klo 9 CRC:n kahvihuoneessa.
Cell Culture Meeting Tue 1.4. CRC coffee room The following was discussed at the meeting 1. An exceptionally busy period in cell culture during February-March has been resolved now. No problems have arisen, and there has been enough space for everyone. 2. Sufficient space has been found for the Cell Locker in the incubator and there has been no need to store it outside. It was agreed that the filters will be regularly replaced when the incubators are sterilized, i.e. during Midsummer and Christmas, and if contamination occurs. 3. The inevitable shortage of space in liquid nitrogen tanks was discussed briefly and it was reminded that it is important for each group to review the cell lines in storage. It would be essential to get in touch with researchers who are no longer actively conducting research and review the samples in their names. The easiest way to start is with electronic lists and thereby find out how much space old samples that are being archived and whose necessity no one knows about (nor whether their ethical permit is still valid) are now taking up. 4. Surgery and Anatomy will take care of the liquid nitrogen tanks from the beginning of July. Sanna has been informed about this. If necessary, an orientation will be organized for the people responsible for the shift during June. 5. The bags containing autoclavable waste from cell culture should be moved with their containers to the cell culture entry room for collection. The bags should be closed using staples and marked as wet or dry waste. Empty containers (with bags) can be found from the cell culture entry room or cold room. The waste will be collected for autoclaving three days a week (Mon, Tue and Wed). If not sufficient enough, then one needs to take the waste to the autoclaving themselves. 6. New gloves and storage boxes have been purchased for the liquid nitrogen tanks. 7. A couple of multichannel pipettes has already been sent for maintenance and calibration, please let Antti know if you notice any more pipettes that need service. 8. Soluviljelijöiden kokous ke 4.6.
CRC:n kahvihuone Kokouksessa keskusteltiin pääosin lähestyvän lomakauden valmistautumiseen liittyen
1. Nestetyppivuoro siirtyy heinäkuun alussa Kirurgialle ja Anatomialle. Tällä hetkellä Ihotaudit hoitaa nestetyppivuoroa. Anja perehdyttää kesäkuussa Kirurgian ja Anatomian vuorosta vastaaville henkilöille (Sanna, Elina, Katja, Minna ja Anni) toimintaperiaatteet.
2. Tiedot soluviljelyyn kesää varten tarvittavista tilauksista (vesi, hanskat, jne) tulee toimittaa Antille viimeistään perjantaina 4.7.
3. Laboratoriotakit viedään pesuun juhannuksen jälkeen maanantaina 23.6. Tarkistakaa takkien taskut siihen mennessä. Soluviljelyn eteishuoneeseen hankitaan keltainen säkki jonne takit kerätään.
4. Inkubaattoreita ryhdytään pesemään yksi kerrallaan, hyllyt voidaan laittaa kuitenkin pesuun useammasta kaapista yhtäaikaisesti. Kesän ajalle jätetään jokaiseen huoneeseen päälle yksi kaappi. Toinen pestään joko ennen tai heti lomien jälkeen riippuen niin työntekijöiden kuin myös välinehuollon aikataulusta. Kaasujen tarkistus voidaan tehdä elokuun alussa kun teknisen puolen henkilöstö palaa lomalta. Cell Lockerin filtterit sovittiin vaihdettavaksi kerran vuodessa kaappien pesun yhteydessä tai tarpeen niin vaatiessa (kontaminaatio). Antti tarkistaa teknisen palvelun kanssa hiilidioksipullojen tilanteen. Jos vaihto on ajankohtainen, huolehditaan se ennen lomakauden alkua. 5. Mykoplasmatestiin on tällä hetkellä listalla viisi näytettä. Seuraava mykoplasmatesti tehdään kesän jälkeen maanantaina 29.9.2025. Merkatkaa jaetussa kansiossa olevaan taulukkoon näytteet jotka haluatte analysoitavan tuolloin.
6. Haravapipettien kalibrointi on tehty. Syksyllä lähetetään porrastetusti Antin koordinoimana myös muut pipetit joiden huollosta on kulunut useampi vuosi tai jotka selvästi ovat huollon/kalibroinnin tarpeessa. Sovittiin säännöllisestä, kerran vuodessa, suoritettavasta tarkistuksesta pipetointitilavuuksiin. Jos niissä huomataan heittoa, silloin on tarpeen lähettää pipetti huoltoon.

1. The liquid nitrogen shift will be transferred to Surgery and Anatomy at the beginning of July. Currently, Dermatology is handling the shift. In June, Anja will familiarize the people responsible for the Surgery and Anatomy shift (Sanna, Elina, Katja, Minna and Anni) with the operating principles.
2. Information about the orders needed for cell culture for the summer (water, gloves, etc.) must be delivered to Antti no later than Friday 4.7.
3. Lab coats will be collected for laundry after Midsummer on Monday, June 23. Please check the pockets of the coats by then. A yellow bag is brought to the entry room of cell culture where the coats will be collected.
4. The incubators will be washed one at a time, although shelves from several incubators can be washed at the same time. One cabinet will be left on in each room during the summer. The others will be washed either before or immediately after the holidays, depending on the schedule of both the employees and the equipment maintenance. CO2 levels can be checked at the beginning of August when the technical staff returns from vacation. It was agreed that the Cell Locker filters will be replaced only once a year when the cabinets are washed or when necessary (contamination). Antti will check the status of the carbon dioxide bottles with the technical service. If the replacement is timely, it will be taken care of before the start of the vacation season.
5. There are currently five samples on the list for the mycoplasma test. The next mycoplasma test will be done after the summer, on Mon 29th Sept. List the samples you bring to the analysis in the shared excel file. 6. The calibration of the multi-channel pipettes has been done. In the fall, other pipettes that have had the maintenance several years ago or that clearly need service/calibration will also be sent in stages, coordinated by Antti. It was agreed that the pipetting volumes will be checked regularly, once a year. If there is a deviation in them, then it is necessary to send the pipette for service.

1. Uusien henkilöiden esittäytyminen. Kerkelän ryhmässä ovat aloittaneet Anja ja Enni sekä Salonurmen ryhmässä Mervi. Sini perehdyttää Anjan ja Ennin ipsilän toimintaan (357B), Saija perehdyttää Merviä joka aloittaa viljelyn laboratoriossa 341B. Uudet henkilöt lisätään laminaarien varausjärjestelmään (Salla), soluviljelijöiden postituslistalle (Reetta) sekä CRC:n yhteisiin kansioihin (IT palvelut, Antti cc:nä). 2. Pipettisettien huollot ovat käynnissä, yksi kerrallaan. Yksi setti huollettu kesän aikana, Pirjo lähettää seuraavan, sen jälkeen lähetetään ipsilän setti. 3. Keskusteltiin pienlaitehankintaehdotuksista. Antti kerää listaa vielä huomiseen keskiviikkoon 3.9. klo 11 saakka. Seuraavat ehdotukset nousivat esille: kolme automaattipipettiä, tarratulostin, valomikroskooppi (korvaamaan nyt huollosta toiseen kiertävää mikroskooppia) jossa olisi nykyaikaisempi kuvausmahdollisuus. 4. Mykoplasmatesti siirtyy näillä näkymin lokakuun loppupuolelle, päivämäärä tarkentuu lokakuun puolivälissä. 5. Nestetyppitankkien tilan rajallisuudesta keskusteltiin. Vanhojen näytteiden kohdalla korostettiin tutkimuslupien voimassaoloa. Eli näytteet, joiden tutkimukseen ei ole enää ole lupaa tulee joko hävittää tai tutkimusluvat tulee uusia. Tästä kannattaa hoksauttaa tutkimusryhmien vetäjiä! 6. Soluviljelylaboratorioiden ylläpitoon liittyen muistutettiin, että laminaarit tulee jättää siisteinä seuraavalle työntekijälle, eli kaikki roskat tulee poistaa laminaarista ja se tulee pyyhkiä kauttaaltaan puhtaaksi. 7. UV-lamppujen tarkistaminen otetaan säännölliseksi rutiiniksi neljä kertaa vuodessa aina lomien kynnyksellä, eli kesä-, syys-, joulu-, ja talvilomien aikaan muistetaan testata lamppujen toimivuus. Merkintä labroissa oleviin seurantakirjanpitoon. Ajastimet on kytketty niin, että lamput on päällä klo 4-5. 8. Sähkökatkoista keskusteltiin, tulevien, minuutin mittaisten ei oleteta vaikuttavan toimintaan CRC:llä mutta kannattaa joka tapauksessa seurata, että kaikki laitteet lähtevät katkosten jälkeen käyntiin ja jatkavat normaalia toimintaa. Sähkökatkoja esiintyy aamuisin klo 6-7 ajalla 15.-22.9. 9. Antti on pois viikon 15.-19.9., kannattaa siis huolehtia ensi viikon aikana Antin kautta soluviljelyyn tarvittavat kulutustarvikkeet ja -aineet. 10. We discussed
1. Introduction of new people. Anja and Enni have started in the Kerkelä group and Mervi in the Salonurmi group. Sini will introduce Anja and Enni to the operation of IPS cell lab (357B), Saija will introduce Mervi who will start working in the lab 341B. New people will be added to the laminar reservation system (Salla), the mailing list (Reetta) and the CRC's shared folders (IT services, Antti as cc). 2. Pipette set maintenance is ongoing, one at a time. One set was fixed during the summer, Pirjo will send the next one, and then the one in IPS cell lab will be sent. 3. We discussed on proposals for small equipment purchases. Antti will collect the list until tomorrow, Wednesday, September 3rd at 11 am. The following proposals were raised: three automatic pipettes, a label printer, a microscope (to replace the microscope that is currently being rotated from one maintenance to another) with more modern imaging capabilities. 4. The mycoplasma test will be moved to the end of October, the date will be confirmed in mid-October. 5. The limited space in the liquid nitrogen tanks was discussed. In the case of old samples, the validity of research permits was emphasized. In other words, samples that are no longer permitted for research should either be destroyed or the research permits should be renewed. It is worth reminding the leaders of the research groups about this! 6. Regarding the maintenance of cell culture laboratories, it was reminded that laminars should be left clean for the next person, i.e. trash bags should be removed from laminars and they should be wiped clean. 7. Checking UV lamps need to be started as a regular routine four times a year, always at the beginning of the holidays, i.e. during the summer, autumn, Christmas and winter holidays. Note in the monitoring records in the labs. The timers are connected so that the lamps are on at 4-5 am. 8. Power outages were discussed, the upcoming, minute-long ones are not expected to affect operations at CRC, but it is worth monitoring in any case that all equipment starts up after the outages and continues normal operation. Power outages occur in the mornings at 6-7 am from 15.-22.9. 9. Antti is away during the week of 15.-19.9., so check the consumables and materials needed at the cell culture and provided by Antti before that. 10. Soluviljelijöiden kokous ti 18.11. CRC:n kahvihuone
1. Uusia henkilöitä aloittaa soluviljelyssä neurologian tiimissä ensi vuoden alussa ja myöhemmin keväällä. Laura perehdyttää heidät toimintaan.
2. Suurempi soluviljelylabra (341B) on jatkuvasti liian kiireinen, vapaata työskentelyaikaa laminaareihin on vaikea löytää etenkin ma, ke ja pe jotka ovat soluviljelyn kannalta olennaisimmat päivät. Pohdimme, kuinka järjestellä ryhmien jakoa eri soluviljelytiloihin niin, että kaikki mahtuisivat joustavasti työajan puitteissa viljelemään soluja. Viereiseen, pienempään 342B labraan mahtuisi yksi tutkimusryhmä lisää, kunhan saisimme järjesteltyä enemmän säilytystilaa. Kokouksen jälkeen kävimme tarkastamassa tilannetta, ja teemme suunnitelmia lisäkaappien/hyllyjen hankkimiseksi.
3. Pohdittiin myös uuden hiilidioksidikaapin tarvetta helpottamaan tilan puutetta 341B tilassa. Sellainen saataisiin mahtumaan tilan ja hiilidioksidiliittymän puolesta, jos tarvittava rahoitus järjestyisi.
4. Nestetyppitankkien tilanpuutteesta myös keskusteltiin. Inventaariota on tehty ja vanhoja soluja heitetty pois. Siitä huolimatta useimmilla ryhmillä tilaa on hyvin rajallisesti, parin laatikon verran. Jotta uusia soluja saadaan mahtumaan, täytyy käytännössä vanhoja heittää pois. Antti vie viestiä eteenpäin laboratoriopäälliköille ja selvittää, olisiko yhteinen arkistointitankki mahdollista järjestää Kiepin 2. kerrokseen soluampulleille, joiden käyttö ei ole aktiivista.
5. Ipsilän eteistilaan asennetaan pimennysverho ke 26.11. Sen jälkeen tilaan asennetaan uusi mikroskooppi (tarkempia tietoja voi kysyä Sallalta), ja vanha tuodaan väliaikaiseen käyttöön 341B labraan jonka vakituinen mikroskooppi on pitkittyneellä ja monivaiheisella korjausmatkalla.
6. UV-lamppujen testaamiseen mietittiin keinoja. Paavo vinkkasi UV testikorteista, joiden saatavuutta selvitellään CityLabin Petran kanssa.
7. Anja tekee mykoplasmatestin ti 25.11. Viimeistään tuona päivänä klo 9 mennessä Anjalle tuodut näytteet ehtivät mukaan.
8. Labratakit viedään pesuun ma 15.12. iltapäivällä ennen joululomaa.
9. Inkubaattoreiden pesujen tarkemmasta aikataulusta ennen ja jälkeen lomaa sovitaan soluviljelijöiden ja välinehuoltajien kanssa.

Cell culture meeting Tue 18 Nov CRC coffee room
1. New staff members will start working in cell culture in the neurology team at the beginning of next year and later in the spring. Laura will familiarize them with the work.
2. The larger cell culture lab (341B) is constantly too busy, and it is difficult to find free working time in the laminar hoods, especially on Mondays, Wednesdays, and Fridays, which are the most important days for cell culture. We are considering how to organize the division of groups into different cell culture rooms so that everyone can flexibly culture cells within their working hours. The smaller 342B lab could accommodate one more research group, as long as we could arrange for more storage space. After the meeting, we went to check the situation and are making plans to purchase additional cabinets/shelves.
3. We also discussed the need for a new incubator to alleviate the lack of space in room 341B. Such an incubator could be accommodated in terms of space and carbon dioxide connection if the necessary funding could be arranged.
4. The lack of space in nitrogen tanks was also discussed. An inventory has been made and old cells have been discarded. Nevertheless, most groups have very limited space, only a couple of boxes. In order to make room for new cells, old ones must be discarded. Antti will pass on the message to the laboratory managers and find out whether it would be possible to arrange a shared storage tank on the second floor of Kieppi for cell vials that are not in active use.
5. A blackout curtain will be installed in the IPS cell lab entrance room on November 26. After that, a new microscope will be installed in the room (more detailed information can be obtained from Salla), and the old one will be brought into temporary use in lab 341B, whose permanent microscope is undergoing a prolonged and multi-stage repair.
6. Ways to test UV lamps were discussed. Paavo suggested UV test cards, the availability of which will be investigated.
7. Anja will perform a mycoplasma test on November 25. Samples brought to Anja by 9 a.m. on that day will be included in the test.
8. The lab coats will be sent for washing on Monday, December 15, in the afternoon before the Christmas break.
9. A more detailed schedule for washing the incubators before and after the holidays will be agreed upon with the cell culture technicians and equipment maintenance staff.
10. The next meeting will be held on Wednesday, January 21, at 9 a.m. in the CRC coffee room.
